DOT1A-mediated histone methylation and replication regulation in Trypanosoma brucei
Histone methylation by DOT1 enzymes is known to regulate various cellular processes in eukaryotes such as gene expression or DNA repair. DOT1A, one of two DOT1 enzymes in Trypanosoma brucei, mediates the mono- and di-methylation of lysine 76 on histone H3 and appears to be a key player in DNA replication initiation. The mechanisms by which this histone mark regulates the replication process are still unknown. My PhD project aims to identify the molecular components involved in this mechanism and the elements that are involved in cell cycle-depemdent regulation of DOT1A activity.
